Abstract：
Beh?et’sdiseaseisasystemicinflammatorydisorderwithrecurrentepisodesoforalulceration,skinlesions,genitalulceration,andintraocularinflammation(uveitis).TheintraocularinflammationisstrictlyassociatedwithTheffectorcells.IL-22isamemberoftheIL-10cytokinefamilythatisinvolvedininflammatoryprocesses.Recently,Th22cellswereidentifiedasaThcellpopulationthatproducesIL-22andTNF-αandaredistinctfromTh1,Th2,andTh17cells.Inthisstudy,weestablishedTh22-typeTcellclonesfromocularsamplestakenfromBeh?et’sdiseasepatientswithactiveuveitis.TheseclonesproducedlargeamountsofIL-22andTNF-αbutnottheTh1cytokineIFN-γandtheTh17cytokineIL-17.CD4+TcellsfromtheperipheralbloodofBeh?et’sdiseasepatientsdifferentiatedintoTh22cellsinthepresenceofIL-6andTNF-αinvitro.ThepolarizedTh22celllinesproducedlargeamountsofIL-22,andthepolarizedTh1andTh17cellsalsoproducedIL-22.Inthepresenceofanti–TNF-α–andanti–IL-6–blockingAbs,Beh?et’sdiseaseTh22-typeTcellsfailedtoproduceIL-22.Inaddition,infliximab-pretreatedTh22cellsandTh22-typeocularTcellsproducedlessIL-22andTNF-α.Moreover,IL-22–producingTcellswereisolatedfrommicewithexperimentalautoimmuneuveitis,ananimalmodelofBeh?et’sdisease,andtheintraocularTcellsfromuveitismodelsproducedlargeamountsofIL-22inthepresenceofretinalAgs.OurresultssuggestthatinflammatorycytokinesIL-22andTNF-αmayplayakeyroleintheocularimmuneresponseinBeh?et’sdisease.
